WHAT is a lysosome?

WHY acidic? Andar ke enzymes acid hydrolases hain — yeh sirf pH 5 ke aaspaas achhe se kaam karte hain. Yeh ek kamaal ka safety design hai: agar lysosome leak kare, toh woh enzymes neutral cytosol (pH 7.2) mein pahunchte hain jahan woh almost inactive ho jaate hain. Toh cell galti se khud ko digest nahi karta.

HOW is the acid maintained? Ek membrane pump jise V-type H⁺-ATPase kehte hain ATP kharchta hai taki protons () ko lysosome ke andar push kar sake:

ko andar pump karne se internal pH kam ho jaata hai (zyaada = zyaada acidic, kyunki ).

WHAT is autophagy?

The pathway (macroautophagy), step by step:

  1. Cytosol mein ek납작 double membrane banti hai jise phagophore kehte hain.
  2. Yeh ek ghise-pite mitochondrion / protein aggregate ke chaaron taraf curve ho jaati hai.
  3. Edges seal ho jaate hain → autophagosome (cargo ko hold karne wali double-membrane vesicle).
  4. Autophagosome ek lysosome se fuse hota hai → autolysosome.
  5. Acid hydrolases cargo ko amino acids, fatty acids, sugars mein digest karte hain.
  6. Yeh building blocks cytosol mein wapas pump ho jaate hain aur reuse hote hain.

WHY does the cell do this?

  • Quality control: damaged organelles ko unke harmful contents leak hone se pehle hata do.
  • Starvation mein survival: jab nutrients khatam ho jaate hain, cell apne khud ke parts kha kar amino acids release karta hai → essential proteins / ATP banata rehta hai.
  • Defense: andar ghuse bacteria ko digest kar sakta hai (xenophagy).

Flashcards

Lysosome kya hai?
Ek membrane-bound organelle jo acid hydrolases se bhara hai aur acidic pH par macromolecules digest karta hai.
Lysosome ka internal pH kitna hota hai?
Lagbhag 4.5–5.0 (acidic).
Lysosomal acidity kaun sa pump maintain karta hai, aur kisse?
V-type H⁺-ATPase, H⁺ pump karne ke liye ATP use karke.
Lysosomal enzymes ko ACID hydrolases kyun kehte hain?
Yeh acidic pH (~5) par optimally kaam karte hain, cytosol ke pH 7.2 par nahi.
Acidic optimum ek safety mechanism ki tarah kaise kaam karta hai?
Agar enzymes neutral cytosol mein leak ho jaayein toh woh almost inactive ho jaate hain, toh cell self-digest nahi hota.
Autophagy define karo.
Cell apne khud ke organelles/cytoplasm ko degrade karta hai unhe ek autophagosome mein bandh karke jo lysosome se fuse hota hai.
Macroautophagy mein cargo ko engulf karne wali vesicle ka naam kya hai?
Autophagosome (double membrane).
Autolysosome kya hai?
Woh structure jo autophagosome ke lysosome se fuse hone par banta hai; digestion yahan hoti hai.
Starvation autophagy kyun badhaata hai?
Kam nutrients mTOR ko inhibit karte hain, autophagy se brake hata dete hain, toh cell fuel ke liye parts recycle karta hai.
Lysosomal storage disease kya hai?
Ek disorder jisme ek missing/defective hydrolase ki wajah se uska substrate accumulate ho jaata hai (jaise, Tay–Sachs).
Lysosome lumen cytosol se kitne guna zyaada acidic hai (pH 4.5 vs 7.2)?
Lagbhag 500× (10^2.7) zyaada H⁺.

Recall Feynman: explain to a 12-year-old

Socho tumhara kamra dheere-dheere toote khilaunon aur puraane khaane se bhar jaata hai. Lysosome ek magic trash bin ki tarah hai jo kachra pighalakar Lego bricks banaa deti hai jinhe tum reuse kar sako. Lekin "pighlaane waala juice" itna strong hai ki woh sirf bin ke andar kaam karta hai (woh khatte acid se bhara hai). Autophagy tum ho jo kamre mein ghoom rahe ho, toote khilaune utha rahe ho, unhe ek bag mein wrap kar rahe ho, aur magic bin mein daal rahe ho. Jab tum bhookhe ho aur khaana nahi hai, tum purani cheezein recycle karke nayi useful cheezein bhi bana sakte ho. Isi tarah cell saaf rehti hai aur survive karti hai.
